How does it know the difference between glycol or DEF or battery water?

Not sure. Sometimes you just have to hit the I believe button.

The measurement scale at left is not labled - is it supposed to be showing g/ml of specific gravity? If so, the top 3 & bottom 3 numbers are wrong.

The picture of this scale is not an enlarged view in proportion. This is a schematic. Thank you for your curiosity and seriousness. But you can't use this diagram to measure any battery fluid.

What type solution should be used to calibrate it? Is there one type or does it depend on what fluid you are going to be testing? Def, anti freeze

Use Refractometer Calibration Fluid. Makes no difference what liquid you are ultimately testing. The Calibration Fluid is a certified standard that results a specific reading.

The refractometer can be used to test glycol concentration and freezing point temperature, as well as other liquid tests. It can be calibrated using Refractometer Calibration Fluid regardless of the liquid being tested. Although the schematic picture of left measurement scale is not labelled for g/ml of specific gravity and no markings on the outside indicate its range or purpose. The device is useful to check if there's no antifreeze in water with a hot water heater.
